<dcterms:abstract xml:lang="fr">Le cancer demeure un défi majeur de santé publique et, malgré les avancées thérapeutiques, les traitements actuels souffrent encore d’un manque de sélectivité et de tolérance. Dans ce contexte, les conjugués anticorps-médicament (ADCs) se sont imposés comme une approche innovante, combinant la spécificité des anticorps à la puissance cytotoxique de petites molécules. Toutefois, leur développement reste limité par la faible diversité des charges utilisées et l’apparition de résistances.Ce travail explore la vérubuline (MPC-6827), un inhibiteur original de la tubuline, comme médicament alternatif pour ADCs. Puissante à de très faibles concentrations, relativement insensible aux mécanismes d’efflux et capable de franchir la barrière hémato-encéphalique, la vérubuline souffre néanmoins d’une instabilité métabolique rapide. Pour y remédier, une stratégie de deutération a été développée, conduisant à des analogues plus stables et actifs. La synthèse, la caractérisation et l’évaluation in vitro et in vivo de ces dérivés deutérés, intégrés à des conjugués au trastuzumab, ont démontré une stabilité accrue, une conservation de l’activité cytotoxique et une efficacité antitumorale comparable à celle d’ADCs de référence. Ces résultats ouvrent la voie à une nouvelle génération d’ADCs plus stables et mieux tolérés, adaptés aux cancers chimiorésistants, contribuant à élargir l’arsenal thérapeutique en oncologie de précision.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Cancer remains a major public health challenge and, despite therapeutic advances, current treatments still suffer from limited selectivity and tolerance. In this context, antibody–drug conjugates (ADCs) have emerged as an innovative strategy, combining the specificity of antibodies with the cytotoxic power of small molecules. However, their development is constrained by the limited diversity of payloads and the emergence of resistance.This work investigates verubulin (MPC-6827), an original tubulin inhibitor, as an alternative payload for ADCs. Active at very low concentrations, relatively insensitive to efflux mechanisms, and capable of crossing the blood–brain barrier, verubulin nevertheless suffers from rapid metabolic instability. To overcome this limitation, a deuteration strategy was developed, leading to more stable and active analogues.The synthesis, characterization, and in vitro and in vivo evaluation of these deuterated derivatives, integrated into trastuzumab-based conjugates, demonstrated improved stability, preserved cytotoxic activity, and antitumor efficacy comparable to reference ADCs. These results pave the way for a new generation of ADCs, more stable and potentially better tolerated, adapted to chemoresistant cancers and contributing to the expansion of precision oncology</dcterms:abstract>
