<dcterms:abstract xml:lang="fr">Mon projet de doctorat vise à développer de nouvelles approches pour lutter contre les infections pulmonaires causées par des bactéries Gram-négatives multirésistantes. Nous ciblons la colistine (CST), antibiotique de dernier recours, dont la toxicité rénale est un enjeu majeur. Pour réduire cette toxicité tout en améliorant son efficacité, nous explorons l'administration par nébulisation afin de diminuer la dose nécessaire. Nos stratégies incluent : 1) l’optimisation de l'efficacité antibiotique via la synergie de molécules naturelles et l’encapsulation dans des nano-émulsions (NEs) ; 2) l’augmentation de la concentration locale du traitement en rendant la CST plus lipophile et en l'encapsulant dans des NEs capables de traverser biofilms et mucus ; 3) la sélection des NEs les plus performantes et stables pour une administration optimisée via inhalateurs courants.</dcterms:abstract>
<dcterms:abstract xml:lang="en">This doctoral research develops innovative strategies against pulmonary infections caused by multidrug-resistant Gram-negative bacteria. It focuses on colistin (CST), a last-resort antibiotic for carbapenem-resistant infections, limited by significant renal toxicity. The key challenge is reducing this toxicity while improving efficacy, enabling lower doses or direct pulmonary administration. Three main approaches are proposed: (1) enhancing antibiotic efficacy via synergistic natural compounds and nanoemulsion (NE)-based delivery; (2) increasing drug concentration at the infection site by modifying CST to a more lipophilic form and encapsulating it in NEs for better stability, biofilm penetration, and mucus diffusion; (3) selecting the most efficient and stable NEs for administration via standard inhalation devices.</dcterms:abstract>
